Abstrakt:
Recent cryo-EM structures of multiple spliceosome forms offer a wealth of information on the arrangement of this impressively complicated ribonucleoprotein complex. Guided by static cryo-EM structures, we use yeast genetics to shed light on the delicate nature of conformational changes at the spliceosomal catalytic center. By modulating stabilities of two competing catalytic and non-catalytic conformations we alter splicing specificity: whereas stabilization of the catalytic conformation allows for splicing of even suboptimal substrates and relaxes splicing specificity, its destabilization results in restrictive, ultra-specific splicing, where only canonical introns can be removed. Understanding mechanisms regulating splicing catalysis will allow us to better predict and modulate alternative splicing patterns.
